As long as you contact them during a time where US citizens are awake, I'd say waiting times are always under 2 hours unless it's about payments. A single email should be enough.
If the site is under maintenance or attack by hackers, replies could take longer - but that's rather rare.
Honestly, you will only get a reply if what you reported on 'requires' a response. Otherwise, they well just deal with the issue and that's it. Have you checked if the issue is already dealt with? (If they determine that there's no issue, then nothing will happen.)
There's also a regular reporting feature (the little [Flag]-button) that might or might not be a solution to whatever you reported on.
Remember that human beings need to read your reports. Be polite.
I contacted support a few times over to e course of 2 months and they never did anything or responded. Tinfoi told me to contact mods and they had it fixed in 10 min! Shout it adr3-n and tomfulp

I contacted a person who can look into it, because that may be a problem a lot of people might have without even knowing. But the rest is out of my hands and they don't exactly owe me an explanation. ;)
